"E" appears at the corner of screen and not able to punch any numbers and beeps until I hit PLU/SUB button an noise goes away. This is happening in Sharp ER-A320 cash register.

I do not know this item, but the easy fix is a power reset. Unplug for 30 sec the replug power. If that doesn’t do it, it may be indicating a drawer jam. other than that, it would be a more serious internal problem.

To make sure it is not a door jam look on the bottom of the register for a lever (small usually metal) and move it to open the drawer. It you move the lever and it does not open it is jammed.
Door is not jammed as I was able to open from the lever on the bottom of the register. I tried to plugging back with JOURNAL and PAPER FEED keys pressed. No luck yet.
