No mic in calls and no speaker function after lightning replacement

EDIT: So I've reassembled the phone and the speaker is now working great, the mic also works great on facetime audio, its crystal clear. The issue seems to be whenever i make an actual phone call instead of a facetime (different aerial?). It works reliably on the phone when using speakerphone but not when i hold it up to my ear and turn speaker off. Very odd.

Had an iPhone 6 with broken lightning port so replaced with new part (the entire flex including headphone jack, and mic). Repair went smoothly with no obvious mistakes made (followed the guide to the letter).

After reassembling and powering on the new lightning port works great however the speaker will not produce sound and people cannot hear me in calls.

The speaker: I have taken the speaker back out of the phone and can see that the pins that connect it to the flex have left indentation on the gold pads, clearly a connection is being made. However I do not notice indentation on the original part. I cannot get ANY sound from the speaker under any function of the phone (yes volume is up).

The mic: The mic doesn't function during voice calls (including on speaker). It initially worked for 1 call but it was described as quiet. What is odd is that i can still use siri (obviously cant hear the response) and it seems to be fine for voice memos too.

Any help is greatly appreciated. I still have the old flex so can use that as a debug tool if needed (only the lightning connector didn't function) .

If both the main mic and bottom loudspeaker not working then it's either the charger port is faulty or the charging port connector is damaged. (You will notice straight away if that was the case.)

Thanks Ben, after reassembly it seems that the speaker and mic work great on wifi calls but I am only having problems with the mic on phone calls.

Ah okay. That sounds like the bottom microphone on the charger port since that is connected on the charging port assembly itself.

I had issues using the replacement charger port without the apple logo on it for iPhone 6.

It was labelled with JBC which I suppose is the supplier name and a obvious sign it's an aftermarket part as it was labelled on the same spot the apple logo would be.

It wouldn't get 4G signal properly after calling when it switched from 3G back to 4G on calls until I replaced the charger port again.

I used one with the apple logo printed on it just to be safe.

When it comes to aftermarket parts sometimes it's better to buy more than one in case it's faulty especially for iPhone replacement parts that are relatively cheap like the charging port for example as there are many various quality parts flowing in the market.

Sorry for the initial confusion. In the medical field they have a saying "When you hear hoofbeats, think horses, not zebras" The first thing you should try is exchanging the part, and hopefully rule out the Lightning dock. After that I would think it's the Cirrus Logic 338S1201 Audio Codec chip. Which requires microsoldering. You'll want to send it off to a board level repair shop

Are you sure? I didn't go near any of those parts and it appears to be the bottom microphone that is not working correctly. Could you explain your answer a bit more?

Sorry, I misread. I'll edit this answer

Ill try replacing the part, likely that the replacement part was faulty (read "It was cheap").
