Replaced Dock and now microphone no longer working

So I had a sound problem with my iPhone 4s. So I ordered a replacement dock connector after doing much research. I carefully followed the guide for tear down and once again to put it all back together.

Powered up my phone and voila! It was repaired. Sound from my speakers again. Or so I thought, until a day or two later when I was trying to make a call, I could clearly hear the other person, but they couldn't hear me. At first I thought it was the other person, but after more in depth testing, I realised it was at my end.

So it appears that my mic no longer works. I took my phone apart 2 additional times to check my connections, rubber boot placement over the mic, made sure the rubber boot was clear of any debris as well as the mesh. My mic still doesn't work. I'm wondering if I got a defective part? Seeing my mic worked fine before the repair, and now it doesn't.

So the repair fixed one problem but caused me another. I can make calls with the speakerphone function, but that isn't always convenient in some locations.

I couldn't tell if there was any plastic that should be removed from the front of the mic unit, but I don't think there is after close inspection.

Really hoping someone has some good input on this matter that can help me resolve this.

Had the exact same thing happen to the wife's 4. After the screen cracked I had it replaced by a local repair guy (who was great...just hadn't stumbled into this issue yet). Once repaired, turns out the symptoms you describe appeared. After a month of speakerphone, I decided she had had enough. So I ripped into it fully expecting to destroy it and replace it with something. Anyway, while reverse engineering the previous repair, I noted that the rubber boot that allows for the proper mic placement had a tiny piece of adhesive cellophane (that I'm sure was supposed to be removed upon install) that had become lodged firmly inside the mic area. It was very small, and almost impossible to see: but there it was. Tweezed that out and voila. Worked like a charm.

I was keyed into something funny because the mic (lower mic of course) would capture any "sound" (i.e. vibration) that was't directed into the mic: hitting it, blowing into hard, etc. Anyway, I know you said you checked for debris perhaps it's worth another look as this thing was small!

I checked the boot twice. And after removing the boot, stuck a small screwdriver in the hole to make sure there was nothing blocking it or debris. I looked at the actual microphone piece and couldn't see any plastic covering it or anything that I could remove. I contacted iFixit and they are sending me a replacement unit. I'm hoping it's just a defective replacement part I got. I'll know more when the new piece arrives and I replace the dock again. Hopefully that will remedy the situation.
